Aerobic Respiration NS201 Ani Galstian Jane Choi James Han Paula Kim Lab 9: Aerobic Respiration Description of Video Our video shows the steps we took to observe aerobic respiration, which consists of the Krebs cycle, and the electron transport chain. We observed this in germinated and dry peas, to see if there is any difference in the amount they respirate. We measured respiration in two ways: by the amount of carbon dioxide being produced, and by the amount of oxygen taken in. In order to measure the amount of oxygen consumed, we added potassium hydroxide (KOH), which absorbs the carbon dioxide being produced during aerobic respiration. Our video shows the amount of respiration undertaken by germinated peas vs. dry peas. Results and Analysis Throughout this lab, we observed the amount of carbon dioxide produced and the amount of oxygen taken in by germinated and dry peas during aerobic respiration. The two parts of aerobic respiration are the Krebs cycle and the Electron Transport Chain, which occur in the Mitochondrion. Carbon dioxide is produced during the Krebs cycle, along with NADH, and FADH2, and some ATP. Afterwards, oxygen is consumed during the electron transport chain. Oxygen is the final acceptor of the chain. When it accepts an electron, the electron binds with a hydrogen ion and the oxygen, forming H2O. Thus, the carbon dioxide production happens before the oxygen consumption. Before measuring our results, we made a few predictions concerning the germinated vs. dry peas. Because dry peas are dormant, we predicted that we would not see evidence of respiration in them. Germinated peas on the other hand are alive, because the enzymes on the outer level of their cells are activated due to the presence of water. Thus, we predicted that we would see clear evidence of respiration. During the next half hour, we measured the respiration going on in the germinated and dry peas. We did this by the movement of the indicator fluid in the respirometers that were attached to the 4 test tubes, two each of germinated and dry peas. When looking at the results of respiration occurring within the germinated peas, we observed that respiration indeed had taken place. 2.4 cm of oxygen was consumed, and 5.4 cm of carbon dioxide was produced. The carbon dioxide production was 3 cm more than the oxygen consumption. This means that photosynthesis was going on in the germinated peas at a much lower rate than respiration. The consumption of carbon dioxide by photosynthesis was less than the production of carbon dioxide production as a result of respiration. This can be due to the fact that the experiment was conducted in dimly lit room. Next, we observed respiration going on in the dry peas. The oxygen consumption was 0.2 cm, and the carbon dioxide production was also 0.2 cm. The fact that these numbers are equal shows that if any respiration was going on during this time was happening at an equal rate as photosynthesis. However, 0.2 cm is a very small number, and could be a result of error. Our indicator fluid in the respirometer could have moved before we started the experiment, which would make our measurement inaccurate. Thus, we could conclude that respiration did not occur at all. RESPIRATION The respiratory system provides oxygen to the body's cells, and removes carbon dioxide Air containing oxygen, enters the body through the nose and mouth, travels through the windpipe, and into the branching airways of the lungs, called bronchi. Here, through thin-walled sacs covered by capillaries, oxygen passes through the lung tissue and goes into the blood stream, where it locks onto red blood cells. Further in the bloodstream, the blood cells squeeze through more capillaries. Here, oxygen unhooks from red blood cells and crosses into other body cells to be used for life-maintaining chemical reactions. I Dream of Aerobic Respiration Our final lab video was a presentation of aerobic respiration, Lab number 9. Aerobic respiration is a process that produces large amounts of ATP in the mitochondrion in the presence of oxygen. The process starts off with pyruvate entering the inner matrix, which is called krebs cyle. During the cycle, pyruvate circles around in the inner matrix and releases ATP and CO2. The pyruvate is changed by a series of reactions to highly reduced electrons called NADH and FADH2. The NADH and FADH2 donate their electrons to the nearby cytochrome. The cytochromes form a chain-like structure on the inner membrane and pass these newly gained electrons down the chain. Making the cytochrome excited and thus pumping protons from the inner matrix into the intermembrane space, through active transport. Chemiosmotic gradient is formed next, this begins when the intermembrane is pH 5 and the inner matrix is pH 7. The ATP synthase becomes activated and forms a channel which allows the built up the protons to pass through the channel, through passive transport, moving back into the inter matrix.
